The clinical trial evidence is form-specific: glucosamine sulfate at 1,500 mg/day shows consistent modest benefit for knee osteoarthritis, while the more common (and cheaper) glucosamine hydrochloride form does not. This distinction explains why so many trials and consumers report different outcomes.
How Glucosamine Sulfate Works
Glucosamine is a substrate for glycosaminoglycan synthesis — the structural matrix of cartilage. Supplementation appears to mildly slow cartilage breakdown, support synovial fluid viscosity, and reduce inflammatory cytokines in the joint. The sulfate form delivers the additional sulfur atom that participates in matrix construction.

Interactions
May enhance the effect of warfarin — monitor INR if you're anticoagulated. Mild interaction with sulfonylureas (theoretical hypoglycemia risk; clinically minor). Generally compatible with most other medications.
